About This Course

This 3-day mentor-based workshop introduces participants to One Health toxicology, environmental contaminant assessment, xenobiotic behavior, toxicity mechanisms, exposure analysis, biomarkers, and computational risk assessment. Participants will gain practical experience using freely accessible toxicology and chemical databases to investigate contaminants, interpret toxicity information, and develop structured risk-assessment workflows.

Aim

To provide participants with an integrated understanding of environmental toxicology, xenobiotic exposure, biological toxicity mechanisms, and computational risk assessment within a One Health framework.

  • Identify biomarkers associated with toxicant exposure
  • Retrieve and interpret toxicity information from public databases
  • Understand environmental and health risk-assessment principles

Workshop Structure

Day 1: Environmental Contaminants, Xenobiotic Fate & One Health Exposure

Focus: Understanding how environmental contaminants move through ecosystems and create exposure across human, animal, and environmental health.

  • Introduction to One Health toxicology and environmental health
  • Major environmental contaminants: heavy metals, pesticides, PFAS, pharmaceuticals, endocrine disruptors, and industrial pollutants
  • Sources, environmental pathways, and routes of toxicant exposure
  • Xenobiotic absorption, distribution, metabolism, and elimination
  • Physicochemical properties influencing environmental behaviour
  • Persistence, transformation, bioaccumulation, and biomagnification
  • Human, animal, wildlife, and food-chain exposure pathways
  • Connecting environmental contamination with One Health consequences

Hands-on Lab: Environmental Contaminant & Exposure Profiling
Participants will select an environmental contaminant and investigate its chemical identity, physicochemical properties, major environmental sources, persistence, environmental fate, exposure routes, bioaccumulation potential, and available human and animal health information using PubChem and the EPA CompTox Chemicals Dashboard.

Hands-on Output: One Health Environmental Contaminant Profile


Day 2: Computational Toxicology, Mechanisms & Biomarkers

Focus: Understanding how toxicants interact with biological systems using mechanistic and computational toxicology evidence.

  • Principles of dose-response relationships
  • Acute, sub-chronic, and chronic toxicity
  • Toxicokinetics and toxicodynamics
  • Cellular and molecular mechanisms of toxicant-induced damage
  • Oxidative stress, inflammation, genotoxicity, and organ toxicity
  • Metabolic activation, detoxification, and susceptibility
  • Biomarkers of exposure, biological effect, and susceptibility
  • Introduction to toxicogenomics, bioactivity screening, adverse outcome pathways, and read-across

Hands-on Lab: Computational Toxicity & Mechanism Mapping
Participants will investigate the selected contaminant to explore toxicity endpoints, associated genes and molecular targets, toxicant–disease relationships, biological pathways, biomarkers, bioactivity evidence, potential adverse outcomes, and related chemicals for read-across interpretation using CTD, EPA CompTox/ToxCast, AOP-Wiki, and GenRA.

Day 3: Exposure Assessment & One Health Risk Characterization

Focus: Translating hazard, exposure, and biological evidence into a structured One Health risk assessment.

  • Fundamentals of environmental and health risk assessment
  • Hazard identification and hazard characterization
  • Exposure assessment for humans, animals, and environmental receptors
  • Understanding LD50, NOAEL, LOAEL, reference dose, and toxicity benchmarks
  • Development of realistic exposure scenarios
  • Estimated daily intake and exposure calculations
  • Hazard quotient, margin of exposure, and safety-margin concepts
  • Risk characterization, uncertainty analysis, and responsible interpretation

Hands-on Lab: One Health Toxicological Risk-Assessment Workflow
Participants will integrate the contaminant profile, environmental fate, exposure pathways, toxicity evidence, molecular mechanisms, biomarker information, human, animal, and ecological evidence, and exposure calculations to prepare a structured risk assessment using EPA CompTox, PubChem, CTD, EPA ECOTOX, and spreadsheet-based risk calculations.
