About This Course

RNA interference (RNAi) is a natural cellular process used to regulate gene expression. This 3-day workshop will delve into the fundamental mechanisms behind RNAi and its application in controlling the expression of specific genes. Participants will explore theoretical aspects of small interfering RNA (siRNA) techniques, from the molecular biology involved to real-world applications in research and medicine.

Throughout the workshop, we will focus on the role of RNAi in biological research, including gene function analysis, therapeutic approaches in diseases like cancer and viral infections, and gene editing techniques. The workshop will offer guidance in designing siRNA experiments, selecting appropriate delivery systems, and interpreting results, making it a valuable tool for molecular biologists, geneticists, and medical researchers.

Aim

This workshop aims to equip participants with a comprehensive understanding of RNA interference (RNAi) and small interfering RNA (siRNA) techniques. It will cover key mechanisms, applications in gene silencing, and how RNAi can be utilized in research and therapeutic strategies. By the end of the workshop, participants will be able to design and implement RNAi experiments for various biological studies.

Day 1: Introduction to RNA Interference (RNAi)

Introduction to RNA Interference:

  • What is RNAi?
  • Historical background and discovery of RNAi (Fire and Mello’s work)
  • Overview of gene silencing pathways
  • Role of small RNAs in gene regulation

Mechanism of RNAi:

  • Step-by-step mechanism (Dicer, RISC complex)
  • Difference between siRNA and miRNA pathways
  • Key proteins involved (Argonaute, Dicer)

Biological Functions of RNAi:

  • Post-transcriptional gene silencing (PTGS)
  • Role in defense against viruses and transposons
  • Applications in gene expression regulation

Applications of RNAi in Research:

  • Functional genomics and gene knockdown
  • Use in model organisms (C. elegans, Drosophila, etc.)
  • Discussion on therapeutics and drug development

Day 2: siRNA Techniques: Design and Delivery

siRNA Basics:

  • What is siRNA? Structure and function
  • Differences between siRNA and miRNA
  • siRNA-mediated gene silencing

Designing siRNAs:

  • Key criteria for designing siRNAs
  • Tools and software for siRNA design (e.g., Invitrogen BLOCK-iT, siDirect)
  • Avoiding off-target effects
  • Selection of target sequences, sense and antisense strand considerations

siRNA Delivery Methods:

  • Lipid-based delivery systems (Lipofectamine)
  • Electroporation and viral vectors (AAV, lentivirus)
  • Nanoparticles for siRNA delivery
  • Challenges in siRNA delivery: Stability, off-target effects, cellular uptake

siRNA Knockdown Assays:

  • Confirming siRNA efficiency
  • Techniques: RT-qPCR, Western blotting, and immunofluorescence

Day 3: Applications of RNAi and siRNA in Research and Therapeutics

RNAi and Disease Research:

  • Role in cancer, neurodegenerative diseases, and viral infections
  • Case studies: siRNA therapies in clinical trials (Patisiran for ATTR amyloidosis)

Therapeutic Applications of siRNA:

  • RNAi-based drugs: Development and FDA approval (Onpattro, Givlaari)
  • Challenges in therapeutic siRNA delivery and stability
  • Ongoing clinical trials using RNAi and siRNA

Gene Therapy and siRNA:

  • Role of siRNA in gene therapy and personalized medicine
  • Ethical considerations and regulatory frameworks

Future Directions in RNAi and siRNA Research:
